CJC-1295 / Ipamorelin: Mechanism and Evidence Review | Flow BioLabs

Research reference, mechanism first: CJC-1295 (no DAC) and Ipamorelin are two growth-hormone secretagogues frequently discussed as a pair. This article explains the mechanism and what the science does and does not establish. It contains no preparation, dosing, or administration instructions.

What CJC-1295 and Ipamorelin Are

CJC-1295 and Ipamorelin are two growth-hormone secretagogues that are frequently paired because they stimulate growth hormone (GH) release through two different, complementary receptor pathways. CJC-1295 (no DAC) is a GHRH analog: it binds the growth-hormone-releasing-hormone receptor on the pituitary and signals it to produce GH. Ipamorelin is a selective ghrelin-receptor (GHSR) agonist: it triggers the actual GH pulse and does so without the appetite stimulation or cortisol and prolactin elevation associated with older, less selective secretagogues. Pharmacology literature describes the pairing as synergistic — one primes production, the other triggers release — while preserving the body’s own pulsatile pattern of GH secretion.

An important distinction in the literature: the “no DAC” version of CJC-1295 has a short half-life (roughly 30 minutes to a few hours) that matches Ipamorelin’s roughly 2-hour half-life, which is why the two are discussed as a pair. CJC-1295 *with* DAC has a much longer half-life (on the order of 6–8 days) and produces sustained, non-pulsatile stimulation — a different tool with a different rationale, not interchangeable with the no-DAC pairing described here. Neither compound is approved for any clinical use.

A Note on Protocols

Flow BioLabs does not publish step-by-step preparation or injection instructions for these compounds — that is a clinical decision that belongs with a licensed provider, not a do-it-yourself recipe.

Reported Benefits — and How Strong the Evidence Is

Research and community reports associate the CJC-1295/Ipamorelin pairing with improved recovery, deeper sleep, and changes in body composition — the downstream effects one would expect from increased pulsatile GH and, in turn, IGF-1. The mechanistic case (two-pathway synergy preserving natural GH pulsatility) is well described in pharmacology literature. The honest framing is that the mechanism is sound and well-characterized, while the specific real-world benefit claims remain under-evidenced in rigorous human trials.

Cautions

Because these are not approved medicines, their regulatory and legal status varies by jurisdiction. Growth-hormone-axis manipulation is not appropriate for everyone — for example, it is generally contraindicated in the context of active malignancy — and candidacy and monitoring are clinical decisions to make with a licensed provider.
