Research reference, mechanism first: 5-Amino-1MQ is an oral compound, not an injectable peptide. This article explains the mechanism and the state of the evidence. It contains no dosing, timing, or cycling protocols — protocol decisions belong with a licensed clinician, not a reference article.
What 5-Amino-1MQ Is (and Isn’t)
5-Amino-1MQ (5-amino-1-methylquinolinium) is a small-molecule quinolinium compound of roughly 159 daltons — not a peptide, despite being sold alongside them by peptide vendors, a source of frequent confusion. It is a selective inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme overexpressed in fat tissue. NNMT methylates nicotinamide (a form of vitamin B3) using S-adenosylmethionine (SAM) as the methyl donor. By inhibiting NNMT, 5-Amino-1MQ is theorized to preserve intracellular NAD+ and SAM levels in adipose tissue, shifting fat-cell metabolism away from storage and toward energy expenditure — notably *without* acting as an appetite suppressant.
A practical consequence of its chemistry: 5-Amino-1MQ has high oral bioavailability (excellent membrane permeability in Caco-2 assays). It is an oral compound and is not formulated for injection.
Reported Benefits — and the State of the Evidence
The interest in 5-Amino-1MQ rests on a coherent preclinical story. In rodent models, blocking NNMT reduced adiposity and improved metabolic markers: Neelakantan and colleagues (*Nature Communications*, 2019) reported that a small-molecule NNMT inhibitor reduced adipocyte size by roughly 30%, increased metabolic rate, and prevented weight gain in high-fat-diet mice without food restriction; Kannt and colleagues (*Scientific Reports*, 2015) and earlier work by Kraus and colleagues (*Nature*, 2014) showed NNMT knockdown improves insulin sensitivity and protects against diet-induced obesity. It is also mechanistically complementary to NAD+ precursors like NMN and NR: those add raw material to the NAD+ pathway, while 5-Amino-1MQ is theorized to reduce a drain on it.
The strength of evidence, stated plainly, is low to moderate and preclinical only. The rodent data are consistent and promising, but no human clinical trial has established efficacy or a validated dose for 5-Amino-1MQ. Reported human protocols derive from allometric scaling and community consensus, not controlled studies.
Cautions
5-Amino-1MQ is a preclinical-only research compound, not an approved medicine. Because human safety data are absent, any decisions about this compound belong with a licensed clinician.
